cultivated in Matrigel to establish an organoid model and subsequently grown on filter inserts, enabling functional assessment in Ussing chamber experiments.In controls, claudin-10 was strongly expressed in acinar canaliculi but absent from ductal TJs, whereas C10b iKOs lacked claudin-10b expression in salivary glands.Saliva volume was decreased (17631 l vs 766 l, p<0.05),Na + (775 mM vs 1255 mM, p<0.01) and Cl -(1023 mM vs 1233 mM, p<0.01) concentrations increased, K + concentration (301 mM vs 391 mM, p<0.01) decreased in three control vs three induced animals (mean SEM, Student's t-test).Organoids expressed Cldn10b, transepithelial resistance ranged from 60 to 700 cm 2 and carbachol elicited transient currents, indicating active Cl -secretion. ConclusionsOur data demonstrate the functional relevance of our models for dissecting the role of claudin-10b in the salivary secretion pathway.
