Plain Language SummaryWhat is this summary about?Lemborexant is medication prescribed for the treatment of insomnia, a disorder where people have trouble falling asleep and/or staying asleep. Lemborexant is a type of medication called a dual orexin-receptor antagonist (DORA). Since lemborexant acts on the central nervous system, it is important to understand whether there is potential for abuse. Abuse potential can be assessed using the United States Controlled Substances Act ‘8-factor analysis’. This review discusses what is known about lemborexant according to each of the 8 factors.What did the researchers find?Lemborexant did not show risk of abuse in nonclinical (animal) testing, but did show risk of abuse in a human abuse potential study. Like other similar medications (other DORAs like suvorexant and daridorexant), people who use sedatives recreationally found lemborexant to be ‘likeable’. However, internet surveillance suggests that DORA drugs have very low abuse-related risks in the community. Further, lemborexant did not produce withdrawal symptoms in phase 3 clinical trials, had no abuse-related side effects (such as euphoria) with approved doses, provided significant benefits on sleep, and didn’t cause next-morning effects (lingering feelings or side effects the morning after taking a sleep medication).What do the findings mean?Like most prescription insomnia medications, lemborexant was placed into Schedule IV as a controlled substance. However, the collective evidence described here suggests that DORAs, including lemborexant, may be less likely to be abused in the real world, thereby having lower risks to public health, than sedative-hypnotic drugs.
