Rituximab vs. Belimumab Treatment in Lupus Nephritis: A Literature Review

Abstract
Monoclonal antibody (mAb) therapy is a promising treatment option in many autoimmune diseases, including systemic lupus erythematosus and its renal manifestation, lupus nephritis (LN). Currently, the only mAb approved for use in LN treatment is belimumab, although rituximab, which has been approved for other conditions, is widely used off-label. Therefore, this literature review seeks to compare the biology of both mAbs and analyze the results of clinical trials performed on them to determine whether rituximab is similar enough to belimumab to be considered for approval. Five randomized controlled trials were analyzed with a focus on the safety and efficacy of each treatment. Although the results for rituximab were insignificant, there are similarities between the mAbs which indicate its viability as a treatment, and there is now increased focus on potential combined treatments using both mAbs.
