Abstract
[RETRACTED] Modified ZYMO DNA/RNA miniprep extraction protocol written in a user friendly way to reduce in-process-decisions and process time. This protocol includes three seperate modifications that were trialled sequentially in consultation with Zymo Research when problem was assumed to be incomplete lysis and clogged filters. Reduced sample mass to reduce clogging of columns and filters. Samples were centrifuged for 45 seconds instead of 30 in first pass through filters as sample only partially passed through Increased DNA/RNA Shield during bead-beating to improve lysis and decrease sample viscosity. Increased Lysis buffer to improve lysis, decrease sample viscosity, and maintain 1:1 ratio with sample slurry-DRShield.
