Abstract
Objective: EHE is a rare vascular sarcoma, characterized by WWTR1-CAMTA1 (90%) or YAP1-TFE3 (10%) gene fusions.VT3989 is a selective TEAD inhibitor that disrupts YAP/TAZ-driven transcriptional activity.This phase I/II study evaluated the safety, tolerability, and preliminary antitumor activity of VT3989 in patients with advanced solid tumors, including EHE.Methods: Patients with locally advanced or metastatic EHE enrolled in the doseescalation and dose-expansion cohorts.Key eligibility criteria included progressive disease (previously treated or treatment-nave), measurable or evaluable disease per RECIST v1.1, and adequate organ function.Confirmation of EHE-associated gene fusions was performed locally using immunohistochemistry, fluorescence in situ hybridization, or sequencing.The primary objective was to assess safety and tolerability.Secondary endpoints included objective response rate, duration of response, and progression-free survival.
