Abstract
This protocol provides a detailed guide for the harvesting, fixation, sectioning, and immunohistochemical staining of dopaminergic (DA) neurons in sections of the substantia nigra of mouse brains, focusing on tyrosine hydroxylase (TH) staining and the counting of the number of DA neurons using unbiased stereological counting methods. The protocol was assembled by members of the PD-AGE network composed of leading experts in the fields of ageing and Parkinson's disease and aims to standardise across laboratories an otherwise commonly used method to ensure reproducibility and reliability in immunohistochemical staining for TH and stereological analyses. This document is intended for researchers involved in comparative studies of mouse models of Parkinson's disease and ageing, with a particular emphasis on assessing dopaminergic neuron integrity in the substantia nigra.
