L'acide bétulinique (3), un composé triterpénique, présente de nombreuses activités thérapeutiques, notamment une activité antitumorale sur les mélanomes humains. De plus, des tests in vivo chez la souris ont démontré que l'acide bétulinique (3) est très peu toxique pour l'organisme faisant de cette molécule un traitement potentiel très prometteur. D a déjà été montré que l'acide bétulinique (3) peut être préparé à partir du bétulinol (2), un composé présent en très forte concentration dans l'écorce du bouleau blanc (Betula papyrifera). \n \nLe premier objectif de cette recherche est donc de développer une méthode de synthèse efficace de l'acide bétulinique (3) à partir du bétulinol (2). La stratégie choisie consiste à oxyder sélectivement l'alcool primaire en aldéhyde pour former le bétulinal (14), puis à oxyder ce dernier en fonction acide sans toucher à la fonction alcool secondaire. La première partie de la synthèse a été réalisée avec un rendement isolé de 35% en utilisant l'oxyde de chrome adsorbé sur silice. L'autre partie de la synthèse, l'oxydation du bétulinal (14) en acide bétulinique (3), est réalisée avec un rendement isolé de 67% en utilisant le permanganate de potassium dans l'acétone à reflux. \n \nLe second objectif du travail est de développer une méthode efficace d'estérification entre un alcool triterpénique et un fragment constitué d'une fonction acide carboxylique. Différents agents activants ont été testés pour condenser l'allobétulinol (18) et l'adipate de monométhyle (17). Le dicyclohexylcarbodiimide (29) et le diméthylaminopyridine (31) ont été utilisés dans le dichlorométhane pour préparer le produit de couplage avec un rendement isolé de 35%. \n
