Le cancer de la prostate est la forme de cancer la plus fréquente chez les hommes au Canada. L’une des approches cliniques explorées pour traiter le cancer de la prostate au stade métastatique est d’effectuer de l’alphathérapie ciblée. Le radionucléide candidat pour ce type de traitement est l’actinium 225 (225Ac) grâce à sa longue demi-vie de 9.9 jours et à sa longue chaîne de désintégration composée de 4 désintégrations alpha et 2 désintégrations β-. Cependant, le développement de radioligands à base d’225Ac comporte certaines limitations, dont la faible capacité de production de ce radionucléide et le nombre limité de chélateurs bifonctionnels pouvant offrir une cinétique rapide et une grande stabilité de complexation à l’225Ac. À ce jour, il existe qu’un seul chélateur marqué à l’225Ac couramment utilisé en clinique qui est le DOTA. Ce chélateur requiert des températures élevées pour le marquage à l’225Ac, ce qui peut entrainer la thermolyse de certains ligands. Le complexe 225Ac/DOTA est aussi instable in vivo. La démétallation du radionucléide entraine une toxicité non désirée par une accumulation de l’225Ac libre dans certains tissus comme le foie et les os. Notre hypothèse est que le développement d’un nouveau chélateur bifonctionnel comportant deux pyridines endocycliques et des groupements fonctionnels hydroxamates pourrait améliorer le marquage et la stabilité de radioligands d’225Ac in vivo. Les objectifs de ma maîtrise sont de synthétiser le chélateur bifonctionnel et d’optimiser les conditions de marquage avec le lanthane, un substitut non radioactif de l’225Ac. Les études de marquage ont été réalisées dans les tampons acétate d’ammonium 0.1M, acétate de sodium 0.1M et HEPES 0.1M, aux pH de 5,5 et 7 et à 25 °C et 50 °C. Le chélateur bifonctionnel a été préparé en 12 étapes avec un rendement global de 9.3%. Le marquage du nouveau chélateur en présence de lanthane peut se faire à température ambiante dans un tampon d’acétate d’ammonium 0.1M à pH de 7. La complexation au lanthane est supérieure à 95% après 3 heures d’incubation. En perspectives, des études de complexation et de stabilité avec l’225Ac et la conjugaison du chélateur bifonctionnel au ligand PSMA se liant fortement à la PSMA surexprimée dans le cancer de la prostate pourraient être les prochaines étapes proposées pour évaluer le potentiel de ce nouveau chélateur pour l’alphathérapie ciblée.
