Over the past several decades, the S. cerevisiae replication system provided excellent tools to study the initiation of DNA replication, and has built basic models to understand this process in higher eukaryotes. Therefore, identification and characterization of budding yeast replication initiation proteins, and their associations with replication origins, have significantly advanced our knowledge of eukaryotic DNA replication. Taking advantage of this tool, I studied the role of the cruciform binding protein CBP/14-3-3 in the regulation of the initiation of DNA replication in the budding yeast. CBP/14-3-3 was previously identified as an origin binding protein, and both its cruciform and origin binding activities are cell cycle regulated. In this thesis, I investigated the two fundamental activities of 14-3-3 proteins involved in DNA replication, namely the cruciform binding activity and the regulation of initiation of DNA replication.
