Tracking androgenic compounds and their effects along a river from wastewater discharge to drinking water uptake

Compounds of Emerging Concern (CECs) are compounds found in the environment and suspected to cause harm but which are not yet regulated.CECs include a wide variety of compounds including pharmaceuticals, antiandrogens, and estrogens.Recently focus has been on endocrine disruption such as estrogenic and antiandrogenic activity but very few studies analyzed antiandrogenic activity of wastewater and surface water.Antiandrogenic compounds are endocrine disruptors suspected to cause health concerns including male reproduction diseases.Wastewater treatment plants are one of the biggest contributors to CECs environmental release.Previous studies have reported CECs and antiandrogenic activity in WWTP effluents and surface water.In the present study, we analyzed the presence, impact and persistence of selected CECs, and specifically antiandrogenic compounds and activities, along a river from a WWTP effluent to a drinking water treatment plant water source.Polar Organic Chemical Integrative Sampler (POCIS) and grab water concentrated using Solid Phase Extraction (SPE) were used to monitor the concentration of CECs in WWTP effluent and surface waters, which are in the ultra trace level.Samples were collected from 8 locations including upstream and downstream of a WWTP, the WWTP effluent and a drinking water source downstream of WWTP.Concentrations of CECs were measured using Liquid chromatography-mass spectrometry and androgenic and antiandrogenic activities were measured using the yeast androgen screen (YAS) assay.Many of the studied compounds were not detected in the WWTP effluent or the river.The majority of the detected compounds in the WWTP effluent, were not detected outside the mixing zone once the WWTP effluent was diluted in the river.Gemfibrozil, ibuprofen, carbamazepine, β-estradiol, atrazine and fluconazole were the CECs still detected further downstream of the WWTP effluent.These compounds were shown to be persistent along the river, which aligns with previous studies indicating that they persist in the environment.17α-Ethinylestradiol, estrone, ibuprofen, sucralose, β-estradiol, gemfibrozil, bisphenol A, nilutamide, androstenedione, triclosan were detected in the river sediment, suggesting that these compounds could be attenuating through sediment sorption.No androgenic activity was detected in the WWTP effluent, river water or sediment samples, while antiandrogenic activity was detected in the WWTP effluent, river water upstream and downstream of WWTP.The WWTP thus had a significant effect on the antiandrogenic activity of the river water and the antiandrogenic activity persisted downstream from the WWTP up to the drinking water source.In addition, The WWTP also contributed significantly to the concentration of carbamazepine and β-Estradiol in the river water.This study underscores the importance of monitoring CECs in the environment since some CECs are present and have shown to be persistent in the environment.In addition, it is important to monitor bioactivity since, as shown in the present study, antiandrogenic activity was detected even though specific antiandrogenic compounds were undetected. RésuméLes contaminants d'intérêt émergent (CIE) sont des composés récemment détectés dans l'environnement et qui sont susceptibles d'avoir des impacts négatifs sur l'environnement et la santé publique mais qui ne sont pas soumises à des normes et règlements.Les CIE comprennent une grande variété de produits dont les pharmaceutiques, les antiandrogènes et les œstrogènes.Récemment, les efforts se sont concentrés sur les effets de perturbation endocrinienne dont l'activité estrogénique et antiandrogénique, mais très peu d'études ont analysé l'activité antiandrogénique de l'eau usée et de surface.Les composés anti-androgènes sont des perturbateurs endocriniens soupçonnés de causer des problèmes de santé, y compris des problèmes de reproduction.Les stations de traitement des eaux usées (STEP) sont l'un des principaux contributeurs aux rejets environnementaux des CIE.Des études antérieures ont confirmé la présence de CIE ainsi qu'une activité anti-androgénique dans les effluents des STEP et les eaux de surface.Dans la présente étude, nous avons analysé la présence, l'impact et la persistance des CIE sélectionnés le long d'une rivière, et plus particulièrement les composés et les activités antiandrogènes, et ce à partir de l'effluent d'une usine de traitement des eaux usées jusqu'à la source d'une usine de traitement d'eau potable en aval.Deux méthodes d'échantillonnage, le POCIS (pour Polar Organic Chemical Integrative Samplers) un échantillonneur passif et l'échantillonnage ponctuel suivi d'une extraction en phase solide, ont été utilisés dans cette étude pour la quantification des CIE présents à des concentrations de l'ordre des ng/L.Des échantillons ont été recueillis à partir de 8 sites, y compris en amont et en aval de la station d'épuration, des effluents de la station d'épuration et de la source d'eau potable en aval de la station d'épuration.Les concentrations de CIE ont été mesurées par chromatographie en phase liquide couplée à la spectrométrie de masse.Les activités androgènes et antiandrogéniques ont été mesurées en utilisant le test de criblage d'androgènes de levure (YAS : Yeast Androgen Screen).Un grand nombre des composés étudiés n'ont pas été détectés dans l'effluent de la station d'épuration ou dans la rivière.Dès la sortie de la zone de mélange entre l'effluent et l'eau de surface, la majorité des composés détectés dans l'effluent de la station d'épuration des eaux usées n'ont pas été détectés étant donné la dilution dans l'eau de rivière.Le gemfibrozil, l'ibuprofène, la carbamazépine, le β-œstradiol, l'atrazine et le fluconazole ont toutefois été détectés en aval de l'effluent de la station d'épuration et se sont révélés persistants le long du cours d'eau, ce qui correspond aux résultats obtenus lors des études antérieures sur la persistance dans l'environnement.17α-Ethinylestradiol, estrone, ibuprofène, sucralose, βestradiol, gemfibrozil, bisphenol A, nilutamide, androstenedione and triclosan ont également été détectés dans les sédiments des rivières, ce qui suggère que ces composés pourraient être atténués par la sorption aux sédiments.Aucune activité androgénique n'a été détectée, que ce soit dans les effluents des STEP ou dans les échantillons d'eau de rivière ou de sédiments, alors qu'une activité antiandrogénique a été détectée dans les effluents des STEP ainsi que dans les cours d'eau en amont et en aval des STEP.La STEP a eu un effet significatif sur l'activité antiandrogénique de l'eau de la rivière et celle-ci a persisté en aval de la STEP jusqu'à la source d'eau potable.De plus, l'usine de traitement des eaux usées a eu également un effet significatif sur la concentration des eaux en carbamazépine et en β-estradiol.Cette étude souligne l'importance du suivi des CIE dans l'environnement puisque certains contaminants sont présents et se sont avérés persistants dans l'environnement.De plus, il est important de surveiller la bioactivité puisque, comme le montre cette étude, l'activité antiandrogénique a été détectée même si des composés anti-androgènes spécifiques n'ont pas été détectés.
