Abstract
Actin microfilaments and microtubules are \nimportant cytoskeletal proteins that regulate endothelial \nrepair through alterations in cell shape and through \nregulation of cell migration following wounding of the \nendothelium. Upstream pathways have been identified in \nthe regulation of actin and microtubule organization, \nespecially small GTPases. Recently, there have been \nnumerous proteins suggested to be capable of regulating \ninteraction between microtubules and microfilaments to \nmediate microtubule regulation of endothelial repair, an \nimportant process in limiting injury to the artery wall \nand in reducing the extent of arterial disease. If \ndisrupted, a rapid repair mechanism is important in \nreestablishing the integrity of the endothelium in order to \nreestablish its function as a macromolecular barrier, a \nthromboresistant surface, and a biologically active \ntissue. Strategies to improve repair should alter the \npathobiology of the atherosclerotic plaque and thus \nimprove the prognosis of patients with atherosclerosis.
