Détection d'effets antimicrobien et antiadhésif d'extraits de coproduits du concombre de mer (cucumaria frondosa) et application dans la lutte contre les biofilms bactériens

Abstract
« Au Québec, l'exploitation de l'espèce de concombre de mer Cucumaria frondosa génère une quantité considérable de coproduits à éliminer. Cette biomasse est reconnue dans la littérature en tant que source de molécules bioactives présentant des activités antimicrobiennes intéressantes, pouvant être valorisées et utilisées dans le domaine des biotechnologies. La présente étude a pour objectif d'évaluer le potentiel antibactérien d'extraits riches en saponines provenant de coproduits de C. frondosa pour des applications dans le domaine agroalimentaire et pour la réduction du biofouling. L'effet antibactérien de ces extraits a été évalué sur des bactéries agroalimentaires et marines, et l'effet antiadhésif a été analysé sur des bactéries marines responsables de formation de biofilms en milieu marin.Les tests d'activité antibactérienne ont portés sur des extraits enrichis en saponine, bruts ou traités par dialyse, préparés à partir de coproduits de concombres de mer pêchés sur la côte nord de la Gaspésie durant l'été 2017. Les analyses ont été effectuées dans des microplaques de 96 puits, en conditions contrôlées, en présence de 5 concentrations d'extraits. Des cinétiques de croissance de 24h ont été réalisées avec huit réplicas du produit par concentration, puis, la densité optique a été mesurée automatiquement toutes les 30 minutes par spectrophotométrie. La cinétique bactérienne a permis de déterminer les temps de latence, la concentration minimale inhibitrice (CMI), les taux de croissance et la densité optique à 24h pour chacune des souches bactériennes en présence des différentes concentrations testées. Une solution pure de saponines commerciales a également été testée dans les mêmes conditions pour vérifier les effets des saponines sur les souches bactériennes.Les résultats obtenus montrent que les extraits expérimentaux (bruts et dialysés) ne présentent pas d'effets antibactériens significatifs par contre, les extraits expérimentaux dialysés présentent des effets antiadhésifs intéressants ; les extraits empêchent l'adhésion des bactéries à partir de la concentration 1mg.mL-1 et on atteint 100% de réduction de l'adhésion à 5mg.mL-1 de concentration. The analyses were performed in 96-well microplates, under controlled conditions, in the presence of 5 concentrations of extracts. Growth kinetics for 24 hours were performed with eight replicates of the product per concentration, then, the optical density was automatically measured every 30 minutes by spectrophotometry. Bacterial kinetics were used to determine lag times, minimum inhibitory concentration (MIC), growth rates and optical density at 24 hours for each bacterial strain in the presence of the different concentrations tested. A pure solution of commercial saponins was also tested under the same conditions to verify the effects of saponins on the bacterial strains.The results obtained show that the experimental extracts (raw and dialyzed) do not show significant antibacterial effects. On the other hand, the experimental dialyzed extracts show interesting anti-adhesive effects; the extracts prevent the adhesion of bacteria from the concentration 1mg.mL-1 and we even reach 100% reduction of adhesion at 5mg.mL-1 of concentration.
