Abstract

Cryptococcus gattii é uma levedura basidiomicética, considerada um patógeno primário e um dos agentes etiológicos da criptococose e foi responsável, recentemente, por uma epidemia de criptococose nas Ilhas Vancouver, Canadá. Dentre os principais fatores relacionados à patogenicidade desse microrganismo estão a cápsula polissacarídica, a produção do pigmento melanina e a capacidade de multiplicação a 37°C, que o torna capaz de estabelecer infecção no hospedeiro. A identificação de proteínas envolvidas na interação patógeno-hospedeiro é de suma importância, possibilitando a compreensão dos mecanismos utilizados pelo patógeno para a infecção. Atualmente poucos trabalhos envolvendo Cryptococcus spp. e análises proteômicas são reportados. Neste estudo a linhagem hipervirulenta R265 de C. gattii foi analisada com o objetivo de evidenciar proteínas diferencialmente expressas no desenvolvimento da levedura a 25°C e 37°C. A análise proteômica comparativa por eletroforese bidimensional possibilitou evidenciar quinze proteínas com expressão aumentada a 25°C e vinte e nove proteínas com expressão aumentada a 37°C. A identificação das proteínas diferencialmente expressas verificadas neste estudo, além de contribuir para um melhor entendimento dos mecanismos utilizados por C. gattii para colonizar o hospedeiro, garantindo seu desenvolvimento na temperatura de infecção, também são potenciais alvos para estudos de desenvolvimento de novas terapias para o tratamento da criptococose ocasionada por este patógeno.
