Assessing toluene biodegradation potential in groundwater using molecular biological tools

Abstract
This study aimed to expand the understanding of microbial mechanisms responsible for toluene degradation within a shallow aquifer. Molecular tools focused on groundwater samples to complement ongoing research at this site. Microbial-facilitated degradation was observed in the groundwater at various depths and locations around the site, constituting a range of locations in non-aqueous phase toluene, the plume, and fringe. Abundance and activity of aerobic and anaerobic degradation pathways were quantified and examined in response to toluene concentrations. Microbial community structure, as identified by amplicon sequence variants through next-generation sequencing, also changed in response to toluene concentrations. Toluene-responsive genera were identified and found to change predominantly at concentrations of approximately 100 mg·L-1. These findings confirm natural attenuation of toluene in groundwater is occurring at this site, contribute to a site conceptual model, and establish a methodological approach that can be used at other sites to quantify microbial degradation in impacted groundwater.
