Abstract
The role of auxin binding protein 1 (ABP1) in the mechanism of action of auxin and of auxinic herbicides was investigated. In 1992, an auxinic herbicide resistant biotype of 'Sinapis arvensis' L. (wild mustard) was identified in a Manitoba field. The precise mechanism of its resistance is unknown, but previous studies suggest a relationship between the binding activity of ABP1, the putative target site, and sensitivity to these herbicides. The focus of this thesis was to express ABP1 from the resistant and susceptible biotypes of wild mustard to evaluate their ability to bind auxin and auxinic herbicides. These ABP1s were expressed using baculovirus (eukaryotic) and GST-fusion protein (prokaryotic) expression systems. Expressed proteins were purified and analyzed for binding to auxin and auxinic herbicides. Various ligand binding assays using affinity columns, size exclusion chromatography and ammonium sulphate precipitation failed to show specific auxin binding by ABP1 from the susceptible biotype.
