Abstract
La Cyclosporine est au coeur des greffes allogéniques de cellules souches hématopoïétiques. Elle est en effet incontournable dans la prévention de leur principale complication : la réaction du greffon contre l'hôte. Mais cette molécule est difficile d'emploi pour les prescripteurs en raison d'une marge thérapeutique étroite et d'une pharmacocinétique extrêmement variable. C'est donc pour optimiser l'utilisation de la Cyclosporine chez ces enfants, en particulier le relais de la voie veineuse à la voie orale, qu'une étude rétrospective été effectuée au CHU pédiatrique Sainte Justine de Montréal et doit paraître dans Bone Marrow Transplantation. Cette thèse se fait alors le préambule de cette publication. Par sa synthèse bibliographique, elle retrace les différents aspects de cette technique. des mécanismes immunologiques inhérents et des thérapeutiques employées en greffes de cellules souches hématopoïétiques.
