Abstract
La polyarthrite rhumatoïde (PR) est une maladie auto-immune touchant environ 1% de la population Canadienne. Les nombreux traitements disponibles sur le marché présentent encore des inconvénients majeurs: résistances aux médicaments sur le long terme, et efficacité partielle ou nulle des traitements. C’est pourquoi il est constamment nécessaire de développer de nouvelles cibles thérapeutiques. À ce propos, l’acide lysophosphatidique (LPA) et la sphingosine-1-phosphate (S1P) semblent jouer un rôle dans l’inflammation chronique impliquée dans l’arthrite. Dans le liquide synovial des patients atteints de la PR, on retrouve non seulement du LPA, mais aussi certains produits de son catabolisme et l’enzyme qui le produit. De plus, le TNF-α associé à l’inflammation est capable d’amplifier les effets du LPA et du S1P. Dans notre étude, on essaie de comprendre les mécanismes utilisés par ces deux lysophospholipides qui pourraient exacerber l’inflammation chronique associée à la PR. Les enzymes qui produisent ces lysophospholipides, ou leurs récepteurs, pourraient être des cibles potentielles dans les futurs traitements de la PR.
