Abstract
Fetal brain injury is a major cause for many long-term neurodevelopmental sequelae after birth.My thesis examines the impact of the clinically asymptomatic systemic fetal inflammation,induced by low-dose lipopolysaccharide (LPS), on brain and gut and the role of cholinergicsignalling in fetal inflammation. I quantified this impact via measurements of the systemicinflammatory cytokines, macrophage and microglia activation in gut and brain, respectively, aswell as by the transcriptome analysis of primary microglia cultures in a well-developed sheepfetus model. My findings indicate that even asymptomatic fetal inflammation triggers brain andgut inflammatory response. I also show that the primary fetal sheep brain microglia LPS doublehitculture induces memory of inflammation potentiating the secondary response to LPS whentriggered again in vitro. In conclusion, a double-hit model was successfully developed to mimicfetal inflammation.
