En 1794, la grande expédition botanique de Nouvelle-Espagne avait exploré la côte Ouest de l'Amérique de Vancouver à Panama, et également Cuba et Porto Rico, mais le Guatemala avait été fortuitement laissé à l'écart. Une mission de deux ans fut confiée à Longinos et à J. M. Mociño.Avec l'aide de la société scientifique locale, les “amigos del país” (prés. Jacobo Villaurrutia), Longinos put installer dans une salle du Palais un triple cabinet d'Histoire naturelle que jouxtaient une pépinière et une salle d'étude. Deux élèves furent formés sur place. La première analyse physico-chimique de l'eau de la capitale fut commentée par les deux savants dans le palais de l'archevêque. L'inauguration du cabinet, le 9 décembre 1796 fut très solennelle
