Abstract
During myocardial infarction, significant myocardial cell loss occurs in a process termed "post-ischemic reperfusion injury", a two-step process where the myocardium first undergoes temporary conditions of ischemia caused by an occlusion of blood flow, followed by the re-establishment of blood flow to the previously hypoxic myocardium. It is believed that the excessive production of reactive oxygen intermediates, such as free radicals and pro-oxidants, during the reperfusion process is a major contributing factor to the observed cell death. In the present study, we sought to investigate the role of the prooxidant, hydrogen peroxide (H2O2), in triggering apoptosis in H9c2(2-1) cells, a clonogenic cell line derived from embryonic rat heart ventricles. We demonstrate that there is a rapid up-regulation of elongation factor alpha1 alpha (EF-1alpha) en route to apoptosis initiated by an oxidative insult in vitro and in vivo, and that this phenomenon may be a necessary early event in the induction of apoptosis.
