The role of Nogo-66 receptor as a potential modulator of endothelial barrier function /

Abstract
The expression of the Nogo-66 receptor (NgR) on different types of endothelial cells was a novel observation alleged by Dr. Bar-Or A. and Dr. Fournier AE (personal communication, April 2004). It was hypothesised that the activation of NgR on endothelial cells could cause an increase in barrier permeability by actin reorganization and tight junction alteration. To verify this hypothesis, primary cultures of human brain endothelial cells (HBECs) as well as the human endothelial cell line EA.hy 926 were exposed to the NgR binding domain of Nogo-A (Nogo-66 peptide). This research demonstrates that endothelial cells are unresponsive to Nogo-66 stimulation. Furthermore and most importantly, the protein expression of NgR in endothelial cells was revisited and it is shown that, contrary to previous beliefs, NgR is absent from HBECs and EA.hy 926. In addition to this new perspective about NgR expression on endothelial cells, this research has yielded potentially relevant data about the effect of bovine serum on tight junction maturation.
