N-cadherin is a prominent cell adhesion molecule involved in developmental morphogenesis and synapse formation. We transgenically overexpressed N-cadherin in a spatially and temporally specific manner using the Cre-loxP system. We overexpressed N-cadherin following the peak developmental period for synaptogenesis using the CamKIIa promoter to explore the effects of N-cadherin upregulation on synapse formation. N-cadherin, in concert with other proteins, plays an important role of adhesion in synapse formation and maintenance. Overexpressing N-cadherin in the hippocampus led to an upregulation of specific components of the synaptic complex; β-catenin, PSD-95, GluN2B and synaptophysin were elevated at the protein level whereas the GABAAR, a marker of inhibitory synapses, was unchanged. Dendritic spine length was reduced in overexpressors while spine diameter and density were indistinguishable from wild-type. These results demonstrate that N-cadherin plays an important role in the regulation of synaptic properties.During development, cell morphogenesis occurs in response to cues that can be permissive or inhibitory. Using microcontact printing we created complex microenvironmental conditions to mimic those observed in vivo to examine their impact on cell growth and differentiation. In this study we used a known growth permissive substrate, polylysine (PLL) and an inhibitory substrate, myelin to examine growth responses of neuronal as well as non-neuronal cells to complex environments. We showed that micropatterning of PLL can be used to direct the adherence and axonal outgrowth of hippocampal and cerebellar neurons as well as other cells such as Oli-neu oligodendrocyte progenitor cell lines and fibroblast-like COS7 cells in culture. By printing myelin in stripes, we were able to show that myelin could impair process outgrowth of Oli-neu cells and that this effect was reversible by using Y-27632 a ROCK inhibitor. Use of micro-engineering techniques provides tools to study the mechanisms underlying diverse cell behaviors on patterned permissive and non-permissive substrates.
