Les gènes ayant une activité biologique en lien avec le gène du récepteur de type 2 de l'interleukine 1 (IL1R2) sont des déterminants génétiques de l'asthme allergique \n \nL'asthme est une maladie chronique des voies respiratoires qui se traduit par une hyperactivité, une obstruction bronchique et un remodelage. Plusieurs facteurs génétiques et environnementaux sous-tendent le développement de cette maladie. En effet, plus de 300 gènes ont été jusqu'à présent associés à l'asthme. Dans ce mémoire, il sera question de certains de ces gènes associés avec les phénotypes reliés à l'asthme et qui sont en lien avec les activités du gène du récepteur de type 2 de l'interleukine 1 (IL1R2). Le gène IL1R2 est différemment exprimé dans les biopsies bronchiques de sujets asthmatiques allergiques comparativement aux sujets témoins et a également été associé avec l'atopie. Un criblage pangénomique (GWAS) a été réalisé sur cinq études d'asthme indépendantes (SLSJ, EGEA, CAPPS, SAGE et BUSSELTON) afin d'identifier des polymorphismes associés aux différents phénotypes reliés à l'asthme. Sept gènes ont été associés: beta-site APPcleaving enzyme-1 (BACE1) et la métalloprotéinase 2 (MMP2) avec l'asthme et l'asthme allergique, spleen focus forming virus proviral integration oncogene (SPII) avec l'atopie et l'asthme atopique, endoplasmic reticulum aminopeptidase (ERAP1) avec l'asthme allergique, interleukin 1 receptor accessory protein (IL1RAP) et interleukin 1 alpha (ILIA) avec l'atopie, et le gène interleukin 1 receptor type I (IL1R1) avec l'asthme, l'atopie et l'asthme allergique. Ces résultats soulignent l'importance du rôle &IL1R2 dans l'asthme et suggèrent qu'il serait pertinent de mieux caractériser sa voie biologique à l'aide d'études génétiques fonctionnelles afin d'avoir une meilleure connaissance du rôle &IL1R2 dans la physiopathologie de l'asthme.
