Selon le Global burden of asthma (2004), l'asthme est une maladie inflammatoire chronique des voies respiratoires des plus fréquentes qui affecte environ 300 millions de personnes dans le monde. Cette maladie touche les individus de tous âges et de toutes conditions, bien que de proportion plus élevée chez les enfants. L'asthme est aujourd'hui considéré, dans l'ensemble de la littérature, comme un trait complexe qui apparaît probablement suite à l'influence de facteurs environnementaux chez des individus génétiquement prédisposés. En raison de la grande hétérogénéité phénotypique et génétique observée dans l'asthme, aucun gène de susceptibilité « majeur » à l'asthme n'est reconnu à ce jour. Les objectifs de cette étude sont donc de mettre en évidence des gènes associés à cette affection ainsi que les interactions gène-gène possibles entre eux, en employant l'approche par gènes candidats. La méthodologie repose sur l'emploi d'une population dite à « effet fondateur » (cohorte originaire du Saguenay-Lac-Saint-Jean) dans le génotypage de ces gènes ainsi que d'une démarche statistique originale basée sur la création de pseudotémoins à partir des trios familiaux de la cohorte dans l'analyse des interactions (effectuée à l'aide du test de déséquilibre de transmission version haplotype, du odd ratio et de l'analyse de régression). Des cinq gènes candidats testés, seulement PPARG a montré une association significative avec l'asthme (p = 0,016) et suggestive avec l'atopie (p = 0,042). Concernant les analyses d'interaction, les trois approches statistiques ont permis d'identifier des interactions potentielles entre les gènes PPARG et IL 13 ou IL4RA et entre les gènes IL4RA et IL4 ou IL1B dans l'asthme, ce qui suggère l'emploi de ce type d'analyse dans la recherche de déterminants génétiques pour l'asthme. \n
