Le nitrate est un problème sérieux dans les systèmes en circuit fermé (l’eau est continuellement recyclée) comme les aquariums, car il s'accumule rapidement en devenant toxique pour certains organismes. La dénitrification biologique est de plus en plus utilisée pour traiter et éliminer le nitrate des eaux usées constituant une alternative efficace et économique. Deux souches bactériennes ont été isolées d’un biofilm issu d’un réacteur de dénitrification à lit fluidisé alimenté au méthanol pour contrôler la concentration de nitrate dans le grand aquarium d’eau marine du Biodôme de Montréal. Ces deux souches sont responsables de l’activité dénitrifiante dans le biofilm. La première souche, Methylophaga nitratireducenticrescens JAM1, est capable de croître en culture pure dans des conditions anoxiques, en réduisant le NO3- en nitrite; Elle ne peut réduire le NO2- en NO. Toutefois, elle peut réduire le NO en N2O, et le N2O en N2. La deuxième souche, Hyphomicrobium nitrativorans NL23, est capable de dénitrification complète, à partir du NO3- jusqu’au N2. Nous avons émis l'hypothèse qu'une complémentarité entre les deux espèces existerait dans le biofilm pour assurer une bonne activité dénitrifiante. Le but du projet a été d'évaluer le degré de cette complémentarité en utilisant une approche de co-cultures. Il a été nécessaire de caractériser la dynamique de l'interaction entre ces deux espèces dans le milieu marin pour établir l'impact et l'apport de chacune à l'intérieur du processus. Très peu d'études ont été faites impliquant des co-cultures de souches bactériennes dénitrifiantes et encore moins en milieu marin. Les résultats obtenus dans ce projet permettront d’améliorer l'efficacité du processus de dénitrification et leur soutenabilité lorsqu'ils seront appliqués à plus grande échelle, ce qui pourra nous amener au développement d'un bioprocédé de dénitrification en milieu marin plus efficace. Avec les connaissances acquises sur les deux souches à travers plusieurs études dans notre laboratoire et avec une série de tests visant à établir les caractéristiques physiologiques et de synergie entre les souches, il a été possible d'établir que la souche JAM1 possède un avantage sur la souche NL23 en ce qui concerne l'assimilation du substrat lorsqu'il est utilisé en co-cultures. La souche JAM1 est capable de réduire le nitrate présent dans le milieu de culture dans un temps plus court que celui observé pour la souche NL23, du fait qu'elle a une affinité plus élevée du nitrate. Cependant, une stimulation préalable de la voie de dénitrification de la souche NL23 a permis à celle-ci de réduire le temps d'assimilation du substrat et ainsi d'améliorer ses performances pour être plus compétente en co-cultures par rapport à la souche JAM1. Les tests d'antagonisme réalisés avec les souches ont montré qu'il n'y a aucun type d'interaction négative entre les souches qui puisse affecter la croissance de l'une d'entre elles lorsqu'elles sont en co-cultures, tout comme aucun type de molécule de communication (quorum sensing) n'est produit qui interfèrerait avec la densité de certaines des souches lors du développement de biofilm. Le suivi des co-cultures a été réalisé aussi bien à l'état planctonique qu'à l'état Biofilm, à travers différentes concentrations de NaCl. À l'état planctonique, la souche NL23 a présenté un taux de dénitrification plus faible lorsqu'elle est trouvée en culture pure par rapport à ce qui a été observé en co-cultures. De plus, la faible capacité de la souche NL23 à croître à des concentrations élevées de NaCl, lorsqu'elle est en suspension a été mise en évidence. Le développement et le suivi d'un biofilm dans un réacteur a donné à la souche NL23 la possibilité de s'acclimater à un milieu marin. La capacité des deux souches à adhérer aux supports a permis la formation d'un biofilm performant, avec une activité dénitrifiante permanente, dans lequel la souche NL23 était toujours active et présente. La souche JAM1 a toujours été trouvée dans une proportion plus élevée à travers les différentes concentrations de NaCl évaluées. Il a été déterminé que la souche JAM1 était capable de produire le soluté compatible appelé de l'ectoïne. Ces solutés compatibles, en plus de maintenir l'équilibre osmotique causée par une concentration élevée de sels ou d'autres solutés non ioniques dans le milieu extracellulaire, ont des propriétés biostabilisantes. L'importance dans cette étude s'est concentrée sur la démonstration que la souche JAM1 est capable de produire d'ectoïne dans le milieu de culture, permettant une meilleure adaptation de la souche NL23 aux changements brusques de concentration en NaCl dans la co-culture. En plus d'établir que la concentration de cette molécule dans le milieu de culture était proportionnelle à l'augmentation de la concentration en sel. La concentration d'ectoïne dans le biofilm du réacteur (co-culture) était plus élevée à des concentrations élevées de NaCl. La présence de cette molécule dans la co-culture biofilm est un facteur important qui expliquerait pourquoi la souche NL23 a été mieux adaptée aux changements de concentration de NaCl dans le biofilm. Ces résultats illustrent la capacité des souches NL23 et JAM1 pour effectuer une dénitrification performante en milieu marin. Bien que la souche JAM1 n'ait pas contribué à de meilleures activités dénitrifiantes spécifiques dans les réacteurs, sa présence était essentielle à la survie de la souche NL23 dans un milieu avec des concentrations supérieures à 1,0% de NaCl. Nitrate is a serious problem in closed loop systems (water is continuously recycled) such as aquariums because it accumulates quickly and becomes toxic to some organisms. Biological denitrification is increasingly used to treat and remove nitrate from wastewater as an efficient and economical alternative. Two bacterial strains were isolated from a biofilm from a methanol-fueled fluidized bed denitrification reactor to control the nitrate concentration in the large marine aquarium at the Biodôme de Montréal. These two strains are responsible for the denitrifying activity in the biofilm. The first strain, Methylophaga nitratireducenticrescens JAM1, is able to grow in pure culture under anoxic conditions, reducing NO3- to nitrite; It cannot reduce NO2- to NO. However, it can reduce NO to N2O, and N2O to N2. The second strain, Hyphomicrobium nitrativorans NL23, is capable of complete denitrification, from NO3- to N2. \nWe hypothesized that a collaboration between the two species must exist in the biofilm to establish a good denitrifying activity. The results obtained in this project will improve the efficiency of the denitrification process and its sustainability when applied on a larger scale, which may lead to the development of a more efficient marine denitrification bioprocess. \nWith the knowledge acquired on the two strains through several studies in the laboratory of Professor Richard Villemur and with a series of tests aimed at establishing the physiological and synergistic characteristics between the strains, it was possible to establish that the JAM1 strain possesses an advantage over strain NL23 with regard to the assimilation of the substrate when it is found in co-cultures. The JAM1 strain is capable of reducing the nitrate present in the culture medium in a shorter time than that observed for the NL23 strain, due to the fact that it has a higher affinity for nitrate. However, prior stimulation of the denitrification pathway of strain NL23 enabled it to reduce the assimilation time of the substrate and thus improve its performance in order to be more competitive in co-cultures compared to the JAM1 strain. \nThe antagonism tests carried out with the strains have shown that there is no type of negative interaction between the strains that can affect the growth of one of them when they are in co-cultures, just as no type of communication molecule (quorum sensing) is produced which would interfere with the density of some of the strains during the development of biofilm. \nThe monitoring of the co-cultures was carried out both in the planktonic state and in the Biofilm state, through different concentrations of NaCl. In the planktonic state, strain NL23 exhibited a lower denitrification rate when it is found in pure culture compared to what was observed in co-cultures. In addition, the low capacity of strain NL23 to grow at high concentrations of NaCl, when it is in suspension has been demonstrated.
