Les examens d'imagerie médicale standards, comme l'échographie, la tomographie par émission de position et l'imagerie par résonance magnétique, permettent d'obtenir des informations physiopathologiques (anatomiques et moléculaires) sur des tissus ou organes d'intérêt. Les informations structurelles sont essentielles pour décrire et traiter plusieurs cas cliniques. Cependant, des biopsies sont souvent requises pour étudier les mécanismes biochimiques qui évoluent conjointement avec les pathologies. Puisque 25 % des biopsies entrainent des complications, il est souhaitable de développer des modalités d'imagerie novatrices. L'imagerie de fluorescence est une modalité en essor qui permet de visualiser de multiples agents d'imagerie simultanément. Le projet propose le développement et la caractérisation de nouvelles sondes fluorescentes qui permettront de détecter l'activité de la myéloperoxydase, une enzyme importante des réactions inflammatoires in vivo. L'application visée consiste en l'imagerie de l'activité de cette enzyme dans le contexte de l'inflammation pulmonaire du rat. Les sondes candidates ont été synthétisées par la plateforme de synthèse de peptides et de sondes d'imagerie de l'Institut de pharmacologie de Sherbrooke. Elles sont initialement peu fluorescentes, mais leur activation par un produit de la myéloperoxydase, l’acide hypochlorique, augmente grandement leur fluorescence. Les sondes mises dans des conditions d'activation devraient voir leur fluorescence augmenter dans le temps jusqu'à activation complète. L’introduction permettra d’approfondir des sujets clés à la compréhension du projet, comme l’évènement inflammatoire pulmonaire visé, les techniques d’imagerie standards et novatrices ainsi que la mécanique d’activation des agents d’imagerie utilisés.
