Abstract

Group II introns are a family of introns found in bacterial and organellar, but not in nuclear genomes. Many group II introns encode proteins termed maturases which are required for their splicing and retrotransposition. Recently four maturase-like proteins were identified in the Arabidopsis thaliana nuclear genome. The proteins encoded by these genes are termed nuclear maturases (NMATs). The four NMAT proteins are classified as NMAT1A, NMAT1B, NMAT2A, and NMAT2B. Nuclear maturases are predicted to have mitochondrial localization signals and a conserved X domain suggesting that these putative maturases are transported to organelles and may function in splicing of group II introns. My results indicate that NMAT1A, NMAT2A, and NMAT2B are all required for embryo viability. Analysis of mitochondrial transcripts from the mutant plants indicate that NMAT2A is necessary for efficient splicing of intron 1 of the nad2 gene, and that NMAT2A is necessary for efficient splicing of intron 1 of the nad5 gene. The results thus confirm the hypothesis that NMATs are required for the specific efficient splicing of mitochondrial transcripts.
