Abstract
Mutations in the Parkin gene cause a juvenile-onset form of Parkinson's disease (PD) that is transmitted in an autosomal recessive fashion. Studies have shown that Parkin mutations account for far more cases of PD than all other known genetic causes combined. Further, mutations have been identified even in apparently sporadic cases, suggesting a broader role. There is now strong evidence that Parkin functions in the ubiquitin proteasome pathway (UPP), the main system for protein degradation in the cell.
