The role of deubiquitinase mysm1 in dendritic cells

Abstract
MYSM1 is a deubiquitinase (DUB) comprising Myb-like, SWIRM, and MPN domains.Previous studies focused on the nuclear function of MYSM1 as the deubiquitinase of histone H2A-K119ub, regulating gene expression in hematopoiesis and immune cell development.Recent studies discovered a novel function of MYSM1 as a negative regulator of the signaling pathways of pattern recognition receptors (PRRs).Two studies by Gekara et al. and one by Wu et al. showed that a cytosolic form of MYSM1 is de novo synthesized in macrophages upon inflammation, followed by subsequent proteasomal degradation.MYSM1 attenuated inflammatory response in these cells, through the removing of K63 poly-ubiquitin from TRAFs, RIP2, and STING downstream of TLR, NLR, and cGAS-STING receptors, respectively.These experiments showed increased production of pro-inflammatory cytokines and type-I interferons in macrophages from Mysm1 -/-and Mysm1 fl/fl LysM-cre mice.Dendritic cells (DCs) play a pivotal role in the immune system, as the antigen presenting cells that connect innate and adaptive immunity.However, the role of MYSM1 in the regulation of PRR signaling in DCs was not previously studied.Here we characterize MYSM1 functions in the regulation of DC responses to inflammatory stimuli, using the bone marrow derived dendritic cell model ( BMDCs) and comparing control and Mysm1-deficient (Mysm1 fl/fl CD11c-cre, Mysm1 fl/fl CreERT2) cells.We further test the role of MYSM1 DUB catalytic activity in DC responses, using DCs that express a catalytically-inactive MYSM1 (Mysm1 D660N ).Our findings establish that the loss of MYSM1 or its DUB catalytic activity at late stages in DCs development in the Mysm1 fl/fl CD11c-cre model does not affect DC cell numbers or their V responses to PRR ligands.In contrast, systemic loss of MYSM1 throughout DC development and differentiation in the Mysm1 Δ/Δ CreERT2 model results in impaired DC development and altered cellular responses to inflammatory stimuli.VI Résumé: MYSM1 est une déubiquitinase (DUB) comprenant des domaines Myb-like, SWIRM et MPN.Précédemment, la recherche était concentrée sur la fonction nucléaire de MYSM1 en tant que déubiquitinase de l'histone H2A-K119ub, régulant l'expression des gènes dans l'hématopoïèse et le développement des cellules immunitaires.Récemment, plusieurs études ont découvert une nouvelle fonction de MYSM1 en tant que régulateur négatif des voies de signalisation des récepteurs de reconnaissance de motifs moléculaires (PRR).Deux études menées par Nelson Gekara ainsi q'une autre menée par Jianguo Wu, ont démontré qu'une forme cytosolique de MYSM1 est synthétisée de novo dans les macrophages lors d'une inflammation, suivie d'une dégradation protéasomique.MYSM1 a atténué la réponse inflammatoire dans ces cellules, par l'élimination de la poly-ubiquitine K63 des TRAF, RIP2 et STING en aval des récepteurs TLR, NLR et cGAS-STING, respectivement.Ces études ont démontré une production accrue de cytokines pro-inflammatoires et d'interférons de type I dans les macrophages des souris Mysm1 -/-et Mysm1 fl/fl LysM-cre.Les cellules dendritiques (CD) jouent un rôle central dans le système immunitaire, en tant que cellules présentatrices d'antigènes qui relient l'immunité innée et adaptative.Cependant, le rôle de MYSM1 dans la régulation de la signalisation PRR dans les CD n'a pas été étudié auparavant.Nous caractérisons ici les fonctions de MYSM1 dans la régulation des réponses des CD aux stimuli inflammatoires, en utilisant des cellules dendritiques dérivées de la moelle osseuse (BMDCs) et en comparant les cellules de type sauvage et celles déficientes en MYSM1(Mysm1 fl/fl CD11c-cre, Mysm1 fl/fl CreERT2).Nous testons également le rôle de l'activité VII catalytique de la DUB MYSM1 dans les réponses des CD, en utilisant des CD qui expriment un MYSM1 catalytiquement inactif (Mysm1 D660N ).Nos résultats établissent que la perte de MYSM1 aux stades tardifs du développement des CD dans le modèle Mysm1 fl/fl CD11c-cre, n'affecte pas les réponses cellulaires aux PRR.Par contre, la perte de MYSM1 du système de la souris durant le développement et la différenciation des CD dans le modèle Mysm1 Δ/Δ CreERT2, entraîne une altération des réponses cellulaires aux stimuli inflammatoires.
