Regulation of Skeletal Muscle Lipid Metabolism by Ghrelin

Abstract
Unacylated ghrelin (UnAG) has an emerging role in the regulation of skeletal muscle lipid metabolism, specifically, as a stimulator of fatty acid oxidation (FAO). The physiological relevance of this has not been determined. UnAG peaks in circulation before mealtimes and decreases rapidly after. Lipids from a meal enter the circulation ~2hrs postprandially. Thus, the temporal relationship of FAO stimulated by UnAG, and the appearance of lipids does not align. We found that UnAG elicited a persisting stimulus on FAO between 2-3hrs after its’ removal, ex vivo. UnAG stimulated an immediate phosphorylation on AMPK, but signalling and increased FAT/CD36 or FABPpm content at the plasma membrane at 2hrs could not explain the increase in FAO. UnAG injection did not affect whole-body substrate utilization. These findings confirm UnAG as a stimulator of FAO and provide evidence that UnAG may influence the handling of postprandial lipids.
