Essential tremor (ET) is the most common, yet the least understood movement disorders.Indeed, despite its prevalence and the daily deleterious effects associated with ET motor symptoms, its pathophysiology remains poorly understood.Recent findings have led to the hypothesis that ET is a syndrome resulting from a neurodegenerative process of the cerebellum.Based on this hypothesis, we focused our study on common proteinopathies and lipid abnormalities in the cerebellar cortex.Postmortem human cerebellar tissues from D rs Ali and Alex Rajput were used in the analysis.Using Western-type immunoblotting we found increased cerebellar levels of TDP-43 C-terminal fragments (TDP-43 CTF) at approximately 35 kDa as well as phosphorylated α-synuclein at serine 129 in ET subjects compared to all non-ET subjects.In addition, TDP-43 CTF were positively associated with the severity of ET.Using chromatographic techniques, analyzes of the cerebellar lipid profiles also showed an increase of dihomo-𝛾-linoleic acid in PC as well as a decrease in total fatty acids (FA), saturated FA (including stearic acid), ω-3 (including docosahexaenoic acid (DHA)) and ω-6 (including arachidonic acid).Moreover, a positive correlation between age of death and DHA levels was present.Therefore, all of these proteomic results support the possible neurodegenerative process occurring in ET cerebellar cortex, while lipidomic results provide new pathological information in addition to proposing a new nutritional therapeutic avenue suggesting that a Mediterranean-type diet could prevent this movement disorder.
