Characterization of a chromate-reducing bacterium, potential for a bioremediation application

Abstract
A Cr(VI)-reducing bacterium, designated CRB5, was isolated from a former wood preservation site contaminated with Cr, Cu and As in Acton, Ontario. CRB5 was able to reduce toxic chromate under aerobic and anaerobic conditions with the formation of amorphous Cr(III) precipitates on the surface layers of the cells. 16s rRNA sequence analysis indicates that it is a pseudomonad sharing close homology to 'Pseudomonas synxantha' but is as of yet an unidentified species. It exhibited a broad range of reduction efficiency in minimal nutrient conditions at temperatures between 4 and 37°C and pH 6 to 9. Reduction was determined to be growth dependent and increased reduction rates were observed with increasing initial biomass density. Experimental results using cell-free extracts indicated that chromate reduction was mediated by a soluble reductase in a manner consistent with a detoxification mechanism. This study provides the basis for assessing the potential of using an indigenous Cr(VI)-reducing strain to reduce and immobilize chromate in a bioremediation application.
