Abstract
La méiose des ovocytes de mammifères débute au stade fœtal puis s’arrête temporairement au stade dictié de la prophase-1. Cet arrêt sera maintenu pendant plusieurs années. In vivo, la méiose des ovocytes ne reprendra que suite au pic de LH (hormone lutéinisante) endogène. Lorsque les ovocytes sont retirés de leur follicule et cultivés in vitro, ils reprennent spontanément leur méiose. Une concentration élevée d’AMPc les maintient en arrêt méiotique caractérisé par le stade de vésicule germinale (GV ou dictié), tandis qu’une diminution de la concentration d’AMPc par l’action des phosphodiestérases (PDEs) permet la reprise méiotique. Cet ouvrage comporte deux volets qui investiguent le rôle joué par deux enzymes, la phosphodiestérase de type 3 et l’AMPK (5’adenosine monophosphate activated protein kinase), dans le mécanisme de la reprise méiotique des complexes-ovocytes-cumulus (COCs) et des ovocytes dénudés (DOs) porcins.
