RÉSUMÉ : Le Nunavik dissimule une diversité moléculaire unique, pourtant peu étudiée. Celle-ci résulte du métabolisme des organismes confrontés aux stress biotiques et abiotiques du climat nordique (froid, rayonnement UV, compétition, etc.). Toutefois, cette richesse moléculaire est actuellement menacée sous l'effet des changements climatiques. En réponse à cette urgence d'étudier la composition chimique du Nord-du-Québec, une étude phytochimique du lichen Stereocaulon paschale a été menée et a conduit à l'isolation de 13 métabolites lichéniques, dont deux dibenzofuranes inédits. Bien que ces deux nouvelles molécules appartiennent à une classe présentant plusieurs activités biologiques d'intérêt, une faible disponibilité par isolation limite leur étude. C'est dans cette optique que notre groupe s'est intéressé au développement d'une voie de synthèse totale donnant accès à ces deux dibenzofuranes inédits, ainsi qu'à quatre analogues, afin de les rendre plus accessibles. De plus, nous voulions par le fait même développer une stratégie de synthèse polyvalente qui ouvrirait la voie à la préparation de plusieurs autres produits naturels appartenant à cette classe. La stratégie de synthèse envisagée est convergente et comprend deux étapes clés. Premièrement, une liaison carbone-carbone a été générée entre deux noyaux aromatiques préalablement fonctionnalisés, via un couplage croisé de Suzuki-Miyaura. Cinq précurseurs monoaromatiques ont été préparés en vue de cette étape, soit trois bromures d'aryle (synthons Ouest) et deux acides arylboroniques (synthons Est). Ensuite, le motif furane a été créé via une réaction d'O-arylation intramoléculaire. Jusqu'à présent, les trois synthons Ouest ont été préparés avec des rendements globaux variant entre 39 et 60 %. Au niveau des synthons Est, l'un d'eux a été préparé avec un rendement global de 46 %, alors que l'autre est disponible commercialement. Le couplage de Suzuki-Miyaura a permis jusqu'à maintenant d'accéder à quatre des six 2-arylphénols ciblés, avec des rendements non optimisés variant entre 15 et 56 %. Finalement, un essai préliminaire pour la cyclisation par O-arylation a permis d'accéder à un premier dibenzofurane avec un rendement de 5 %. En bref, nous sommes en mesure d'affirmer que l'approche mise de l'avant s'applique bel et bien à la synthèse de dibenzofuranes complexes. This results from the metabolic response of organisms confronted with northern climate biotic and abiotic stress (cold, UV radiation, competition, etc.). However, this molecular richness is now threatened by the effect of climate change. In response to this emergency to study the chemical composition of the Nord-du-Québec, a phytochemical study of the lichen Stereocaulon paschale was conducted and led to the isolation of 13 lichen metabolites, two of which were new dibenzofurans. Although these compounds showed some bioactivity in preliminary assays, their low abundance limited their study. In order to solve this abundance issue, our group decided to work on the total synthesis of those two dibenzofurans natural products, as well as four other analogues. In a way to allow an easier general access to this class of molecules, we also wished to design a versatile synthetic route towards heavily functionalized dibenzofurans. The planned synthetic strategy is convergent and involves two key steps. First, a carbon-carbon bond was generated between two previously functionalized aromatic rings via a Suzuki-Miyaura cross-coupling. Five aromatic precursors must be prepared for this step: three aryl bromides (West synthons) and two arylboronic acids (East synthons). Afterwards, the furan moiety will be created via an intramolecular O-arylation reaction. So far, the three aryl bromides have been prepared with overall yields between 39 and 60 %. For the two arylboronic acids, one has been prepared with an overall yield of 46 %, while the second one is commercially available. Suzuki-Miyaura cross-coupling provided access to four out of the six 2-arylphenols with an unoptimized yield between 15 and 56 %. Finally, a preliminary experiment with the O-arylation cyclization reaction provided access to a first dibenzofuran with a yield of 5 %. In summary, we can affirm that our proposed approach does lead to complex dibenzofuran synthesis.
