Associations between specific bovine leukocyte antigen DRB3 alleles and mastitis in Canadian Holsteins

Abstract
Associations between bovine leukoctye antigen alleles 'DRB3.2*16 ' and '*23' and mastitis in Canadian Holsteins were investigated. In order to facilitate the genotyping of a large population, a rapid PCR screening method, multi-primer target-PCR (MPT-PCR) was developed. Four primers were combined in one PCR reaction to detect the presence of alleles ' DRB3.2*16' and '*23' simultaneously. Cows (n = 3157), from 56 herds monitored for mastitis for 18 months, were then genotyped by MPT-PCR. No associations were found between the alleles and clinical mastitis, mastitis at calving or somatic cell score. However, 'DRB3.2*23' was significantly associated with an increased occurrence of subclinical mastitis caused by bacterial pathogens (p <= 0.10), and specifically by ' Escherichia coli' (p <= 0.10) and by 'Streptococcus' spp. (p <= 0.05). In contrast, 'DRB3.2*16' was associated with a decreased occurrence of subclinical mastitis caused by 'Strep. ' spp. (p <= 0.05). Further studies are required to reveal the underlying biological mechanisms contributing to the increased or decreased risk of disease associated with these alleles.
