Arsenic is a well-known carcinogen, but has also proved useful as a therapeutic agent. Administration of inorganic arsenic trioxide (ATO) to acute promyelocytic leukemia (APL) patients represents its best known usage as an anti-cancer agent. However, there is less evidence in the literature about the ability of other cancers to respond equally well to arsenic at clinically achievable doses. This thesis reports results obtained by testing a new organic arsenical, S-dimethylarsino-glutathione (ZIO-101) in various hematologic malignant cell lines in vitro. This novel compound appears to be more potent than ATO at mediating apoptosis in APL cell line NB4, in multiple myeloma cell line IM9, as well as in acute lymphoblastic leukemia cell line CCRF-CEM, in vitro. ZIO-101 is also able to induce apoptosis in an ATO-resistant cell line derived from NB4 cells, AR2. These data provide the first in vitro evidence that the organical arsenical ZIO-101 might have a broader thrapeutic spectrum than ATO.
