Cytotoxicité de l'isocaryophyllène sur les cellules L-929 : caractérisation du mécanisme d'action

Abstract
L'isocaryophyllène, un sesquiterpène d'origine végétale, s'est révélé toxique contre plusieurs lignées de cellules cancéreuses tout en étant significativement moins toxique envers les cellules saines. Cette molécule pourrait, par conséquent, être éventuellement intégrée à un traitement anticancéreux. Toutefois, son mécanisme d'action est inconnu. \n \nLe projet rapporté dans le présent mémoire a pour but de caractériser le mécanisme d'action de l'isocary ophyllène. Pour atteindre cet objectif, les effets de l'isocary ophyllène sur une lignée de cellules tumorigènes, les cellules L-929 (fibrosarcomes murins), ont été étudiés. L'administration d'isocaryophyllène à ces cellules s'est accompagnée d'une génération d'espèces oxygénées réactives (ROS). Une peroxydation des lipides membranaires et une perméabilisation de la membrane plasmique ont aussi été observées. La protection des membranes plasmiques avec un antioxydant liposoluble, l'a-tocophérol, a diminué la cytotoxicité de l'isocaryophyllène. À l'inverse, la protection avec un antioxydant hydrosoluble, le trolox, s'est révélé sans effet. Aucune implication du glutathion (GSH) et de la chaîne de transport des électrons n'a été observée dans le processus. Toutefois, certains indices permettraient d'envisager la possibilité que l'isocaryophyllène est oxydé par l'anion superoxyde pour former un époxyde. Cet époxyde pourrait avoir un rôle dans la toxicité de la molécule. \n
