Abstract
Plant responses to temperature extremes are complex and involve multiple regulatory mechanisms.Transcriptional regulation is a major aspect involved in the acclimation and acquisition of tolerance to temperature extremes.The epigenetic landscape of stress-responsive gene promoters influences gene expression concomitantly to the action of transcription factors.Here, the role of histone acetylation, mediated by the SAGA complex's enzymatic subunit Gcn5, was assessed in the monocot model species Brachypodium distachyon using validated inhibitor treatments and Gcn5 overexpression lines.The use of these tools indicated that transcriptional regulation of both cold-and heatresponsive gene expression is influenced by Gcn5-mediated histone acetylation.In addition, two Gcn5 splice variants that have recently been identified in B. distachyon have been functionally characterized.These alternative transcripts differ based on the presence and integrity of conserved domain sequences.L-Gcn5 is missing the bromodomain, which
