Introduction : Les maladies cardiovasculaires (MCV) représentent une cause importante de mortalité et de morbidité dans le monde, et ce, malgré les traitements disponibles. L’athérosclérose qui est un processus immuno-inflammatoire sous-tend ces maladies. L’un de ces principaux acteurs de cette inflammation sont les monocytes par le changement de leur phénotype et de la production incontrôlée de cytokines proinflammatoires. Objectif : L’objectif principal de notre étude est d’investiguer le potentiel nutraceutique de l’huile d’olive extra vierge (HOEV) sur l’état inflammatoire monocytaire chez des participants présentant différents niveaux de risque pour les MCV. Méthodes : 29 participants âgés de 65 ans et plus ont été recrutés, dont 5 personnes âgées en bonne santé, 10 patients avec hypercholestérolémie nouvellement diagnostiquée et 14 patients postinfarctus (3 mois après la survenue de l’infarctus). Les participants des 3 groupes ont été supplémentés en HOEV (25 ml /jour) durant 6 mois. Les prises de sang ont été effectuées après un jeûne de 12 h. Les monocytes ont été isolés à l’aide la technique de Ficoll paque et Percoll. Le phénotype monocytaire et la production de cytokines IL-1β, IL-6, IL-10, TNF- α au repos et après 4h de stimulation par lipopolysaccharide (LPS) ont été déterminé par la cytométrie en flux. Résultats : À l’état basal (pré intervention), les patients dyslipidémiques, en comparaison aux participants en santé ou post-infarctus, ont des monocytes caractérisés par un état inflammatoire de par leur faible production d’IL-10. Nos résultats indiquent que le phénotype monocytaire ainsi que la production d’IL-10, IL-6 et TNF-α ne sont pas modulées de façon significative par l’intervention d’HOEV dans aucun des groupes, que ce soit au repos ou après stimulation par le LPS. Cependant la production d’IL-1β est significativement modulée par la supplémentation en HOEV, plus spécifiquement une augmentation de l’IL-1β chez les patients dyslipidémiques et post Infarctus. Paradoxalement, après 6 mois de supplémentation en HOEV, les monocytes provenant de patients dyslipidémiques stimulés au LPS produisent moins d’IL-1β, et ce en dépit d’une augmentation d’IL-1β à l’état basal. Conclusion : Nos résultats indiquent que l’intervention avec l’HOEV n’a pas induit de changement des phénotypes monocytaires, cependant cette intervention a eu un effet modulatoire sur la production des cytokines.
