Abstract

With the objective to construct single chain variable fragments (scFv) with enhanced ability to neutralize bovine herpes virus-1 (BoHV-1), V[lambda]J[lambda], and VHDHJ H recombinations isolated from anti-BoHV-1 IgG1 secreting hybridoma, were linked together by two amino acids. The V[lambda]J[lambda] -linker-VHDHJH amplicon was cloned into pPICZ[alpha] vector for expression in 'Pichia pastoris'. A recombinant scFv (scFv5-2L), secreted by 'P. pastoris' was characterized. The scFv5-2L had significantly higher BoHV-1 neutralization kinetics (p<0.001) 'in vitro' when compared to a scFv with an 18 amino acid linker (scFv3-18L). Such increased virus neutralization by scFv5-2L is likely due to higher avidity as a result of multimerization per se. The recombinant scFv recognized a viral neutralizing epitope on a 32 kDa envelope glycoprotein BoHV-1. In conclusion, the recombination scFv5-2L against BoHV-1 provides an improved potential diagnostic and therapeutic agent for the prevention and eradication of an economically important viral disease of cattle.
