Au cours de la cryoconservation, le spermatozoïde subit d'importants dommages. Ces dommages prennent la forme de perte de motilité ainsi que de dommages sous-létaux à différents niveaux. Parmi ces derniers certains effets cryo-capacitants sont de plus en plus documentés. Ils contribuent à diminuer la durée de vie et à réduire le pouvoir fécondant du spermatozoïde cryoconservé. Au cours de cette étude, nous avons étudié le comportement de quatre marqueurs spécifiques de compartiments subcellulaires différents suite à la cryoconservation du sperme humain. La protéine P34H, l'a-tubuline, l'acrosine et les phosphotyrosines représentaient respectivement, la membrane plasmique, le cytosquelette, la matrice acrosomale (via l'entrée de calcium) et les voies de signalisation intracellulaires. Chaque marqueur a été étudié avant et après cryoconservation. Une perte de 50% de la protéine P34H, une augmentation de détection de 200% de l'a-tubuline, une activation de la pro-acrosine en sa forme active, la p-acrosine, et une augmentation de la phosphorylation de deux protéines en leurs résidus tyrosine ont été observées. De plus, l'augmentation de détection de l'a-tubuline s'est révélée proportionnelle au temps d'entreposage dans l'azote liquide, une différence significative étant présente dès 15 semaines d'entreposage. À la lumière de ces résultats, il apparaît évident que la cryoconservation du sperme humain est associée à des cryo-dommages sous-létaux prenant place dans différents compartiments de cette cellule hautement spécialisée. Ces dommages contribuent certainement à la perte de pouvoir fécondant observée dans le sperme cryoconservé.
