Kaspaasit ovat ryhmä proteaaseja, jotka pilkkovat kohdeproteiinejaan tietyn aminohapon kohdalta. Niillä on aktiivisessa keskuksessaan kysteiiniä sisältävä sivuketju, joka toimii nukleofiilinä peptidisidoksen hydrolyysissä. Kaspaasit osallistuvat muun muassa solujen tasapainon ylläpitoon, ja niitä pidetäänkin apoptoosin biomarkkereina. Apoptoottisten kaspaasien lisäksi on olemassa myös tulehduksellisia kaspaaseja. Monet sairaudet liittyvät kaspaasien ja apoptoosin epänormaaliin toimintaan. Siksi kaspaasit ovatkin potentiaalinen lääkekohde, ja niille on kehitetty erilaisia peptidi-, pienmolekyyli- ja allosteerisia inhibiittoreita. \nMonet fluoresenssiin perustuvat kaspaasien aktiivisuuden havainnointiin käytettävät menetelmät hyödyntävät Försterin resonanssienergiansiirtoa (FRET), jossa energiaa siirtyy säteilemättömästi virittyneeltä donorilta akseptorille. Energiansiirto tapahtuu dipoli-dipolivuorovaikutusten avulla, kun donori ja akseptori ovat riittävän lähellä toisiaan, ja kun donorin emissiospektri on osittain päällekkäinen akseptorin absorptiospektrin kanssa. Kaspaasien havainnointiin käytettävät FRET koettimet sisältävät tietylle kaspaasille spesifin peptidisekvenssin, johon on konjugoitu donori- ja akseptorimolekyylit vastakkaisiin päihin. Kaspaasin pilkkoessa peptidin donorin ja akseptorin välinen etäisyys kasvaa, mikä voidaan havainta joko donorin fluoresenssin kasvuna tai akseptorin fluoresenssin laskuna. \nKaspaasien havainnointiin voidaan soveltaa myös aggregaation aiheuttamaa emissiota (AIE), jossa fluoresoivan molekyylin kasautuminen aiheuttaa fluoresenssin kasvua. AIE-koettimessa on peptidi, jonka proteaasi voi tunnistaa ja pilkkoa, mikä johtaa lopulta AIE-molekyylien aggregaatioon ja siten fluoresenssiin. Lisäksi kaspaaseja voidaan havainnoida fluorokromeilla leimatuilla inhibiittoreilla, jotka sitoutuvat suoraan aktivoituneen kaspaasin aktiiviseen keskukseen.
