Glioblastoma multiforme is a devastating disease for which available treatments consistently fail.The current clinical standard, temozolomide, is ineffective in most patients due to resistance mechanisms such as the upregulation of DNA repair enzymes.The main objective of this thesis is to report on pharmacological and nanotherapeutic modulators of glioblastoma, focusing on 1) the mechanism of action of a novel histone deacetylase inhibitor (sahaquine), 2) combination treatments targeting tumor defenses and 3) effects on microglia and astrocytes in the tumor microenvironment.Histone deacetylase 6 inhibitors are promising therapeutic agents for glioblastoma due to their effect on cytoplasmic targets (cytoskeleton, kinase signalling) important for cancer invasiveness and growth.We explored the mechanisms of sahaquine, a novel hybrid molecule combining two FDA-approved drugs: primaquine and vorinostat.We showed sahaquine to be List of Abbreviations
