Uzun ve sağlıklı yaşamın, insan genetiği ile ilgili olduğu eskiden beri bilinen bir gerçektir. Uzun yaşayan insan populasyonlarının belirli coğrafi bölgelerde ve bazı etnik gruplarda yoğunlaştığı araştırmalarda yer almaktadır. Her ne kadar uzun yaşam beslenme, sigara ve alkol kullanımı, kişinin çalışma koşulları, iklimsel etkenler, çevre kirliliği, sedanter yaşam, obezite ve stres gibi birçok koşuldan etkileniyorsa da uzun yaşamın genetik kökenli olduğu bilimsel olarak birçok kez kanıtlanmıştır. Moleküler Biyoloji’deki gelişmelerle birlikte bunun detayları da araştırılmaya başlanmıştır.\nKalp hastaları için önemli olduğu düşünülen ACE, eNOS ve PON 1 genlerinin uzun yaşamla da ilgili olabileceği düşünülmektedir. Kalp hastalığı ve kanser toplumda en çok ölümlere neden olan hastalık çeşitleridir. Bunları etkileyen genetik faktörlerin uzun yaşamı da etkileyebileceği düşünülmektedir. Uzun yaşamla ilgili olarak pek çok farklı genetik faktörler gösterilmektedir. Biz kendi şartlarımızda ACE, e-NOS ve PON 1 genlerini çevremizde bulabildiğimiz 90 yaşını geçmiş sağlıklı kişilerde tarayarak sonuçlarını genç populasyonla karşılaştırdık. Bu amaçla 90 yaşını geçmiş sağlıklı 33 kişi ile anket yaparak, kendi istekleri doğrultusunda araştırmamıza katılmalarını sağladık. Bu bireylerden alınan kan örneklerinden DNA izole edildi ve bu DNA örnekleri polimorfizm tayinlerinde kullanıldı. Kontrol grubu olarak; 18- 40 yaş arasında ACE için 48, PON1 için 26 ve eNOS için 50 bireyden oluşan kan örnekleriyle çalışıldı. Sonuçlar SPSS istatistik programıyla anlamlılık derecesi p<0.05 kriteri ölçüt alınarak değerlendirildi. Çıkan sonuçlara göre uzun yaşam ile ACE (I/I, I/D, D/D) gen polimorfizmi arasında anlamlı bir ilişki bulunurken, e-NOS –786 C (T/T, T/C, T/T) ve PON 1 192 (Q/R, R/R, Q/Q ) polimorfizmleriyle anlamlı bir ilişki bulunamamıştır.\n SUMMARY\n It is known that long and healty life has been associated with human genetics since the old times. It is a fact that, in some geographical areas or ethnic groups there are populations living longer than the other people in the other areas of the world. Although having a long life is affected by the use cigarette and alcohol, working conditions, environmental pollution, regulative actions, obesity and stress condition, it is dependent on the genetic origin and that has been proved by scientific research several times. \nThe details of this fact have been searched with the developments in molecular biology. It is thought that ACE, eNOS and PON1 genes that are important for heart patients are also related with having a long life. Heart disease and cancer are the most common causes of death in the world. It’s believed that the genetic factors affecting these illnesses are related to having a long life. We scanned the ACE, e-NOS and PON1 genes of the healthy people over 90 years and compared the results with that of the young generation.\nWith this aim, we made inquiries with 33 healthy people who are over 90. We took their blood samples under doctor control and we isolated DNA from these blood samples and used DNA samples for polymorphism studies. As the controls, the blood samples taken from 18-40 years old people were studied. The results were calculated with SPSS statistic programs. At the end, we didn’t find any significant associations with eNOS and PON 1 192. We found a significant association between ACE and longevity.
