<br>Les lymphocytes T sont des composantes importantes de la réponse immunitaire adaptative dans
\nl'élimination des différentes infections. Dans le cas d'infections virales, cette élimination se fait via
\nla production de cytokines antivirales par les lymphocytes T CD4 (T-CD4s) helper pour activer
\nles différentes populations immunitaires nécessaires, notamment les lymphocytes T CDS (TCDBs)
\nqui sont aussi connues pour leur action cytolytique menant directement à la lyse de la
\ncellule infectée.</br> <br>
\nAfin d'assurer des réponses antivirales et cytotoxiques optimales, les lymphocytes nécessitent
\nun apport énergétique important pour proliférer et exprimer plusieurs cytokines simultanément,
\nd'où le terme polyfonctionnalité. Cet apport énergétique est assuré par l'oxydation de plusieurs
\nsubstrats carbonés menant à la génération de l'énergie sous forme des molécules d'Adénosine
\nTriphosphate (ATP). une plasticité métabolique permettant aux cellules d'altérer leurs voies
\nmétaboliques afin de dégrader différents métabolites et les transformer en ATP. Plusieurs études,
\ndont celles menées dans notre laboratoire montrent que l'une des voies les plus utilisées par les
\nlymphocytes T activées est la beta oxydation des acides gras (FAO).</br> <br>
\nPlusieurs substances peuvent inhiber les réponses immunitaires cellulaires, telles que les
\ncannabinoïdes , dont la consommation est légalisée au Canada. La consommation de cannabis
\npar la population générale est principalement du cannabis à spectre complet avec soit une teneur
\nélevée en cannabidiol (CBD), soit une teneur élevée en tétrahydrocannabinol (THC). L'effet du
\nCBD est plutôt thérapeutique vu qu'il est connu pour son effet anti inflammatoire et anti-douleurs.
\nCependant, les consommateurs de Cannabis à titre récréationel recherchent un effet
\nneurotropique auquel on réfère par le terme 'High ', qui est trouvé typiquement en consommant
\ndu cannabis à teneur élevée en THC. Cette consommation , ayant augmenté depuis la légalisation
\ndu cannabis, et d'autant plus depuis le début de la pandémie actuelle afin de réduire le stress
\nmental dû aux conditions d'isolement et de distanciation imposées au Québec.</br> <br>
\nDans le cadre de la nécessité de la génération d'une réponse immunitaire suffisante et protectrice
\ncontre le SRAS-CoV-2 suite à la vaccination, on se demande si cette consommation de THC ne
\nserait pas de nature inhibante quant à la réponse vaccinale des consommateurs récréationels,
\nqui représentent une population assez importante au Québec et au Canada.</br> <br>
\nDe ce fait, on s'est intéressé à l'étude de l'effet du THC sur les réponses antivirales des
\nlymphocytes T-CDBs et des T-CD4s mémoires.</br> <br>Cette étude a été principalement réalisée en utilisant la plateforme de cytométrie en flux nous
\npermettant de cibler à partir des cellules mononuclées du sang périphérique (CMSPs), nos
\ncellules et cytokines d'intérêt, étant respectivement les lymphocytes T-CD8s et l'IFNy, la TNFa
\nainsi que l'IL -2, via un cocktail multiparamétrique d'anticorps. Nos résultats ont confirmé que les
\nlymphocytes T-CD8s et T-CD4s mémoires, activées de manière polyclonale et, traitées ou non ,
\nau THC pendant 24h, présentent une inhibition significative de leur polyfonctionnalité immunitaire.
\nCes cellules présentent aussi une inhibition de leur potentiel cytotoxique observée via leur
\nexpression des molécules de la cytotoxicité cellulaire, Granzyme B et Pe1io1·ine . De façon
\nimportante, on démontre une possible atteinte du métabolisme lipidique, en évaluant l'expression
\nintra-mitochondriale du transporteur lipidique CPT1 a, qui se trouve significativement diminuée
\nchez les lymphocytes T-CD8s activés en présence de THC. Ces données métaboliques primaires
\nsont à confirmer avec des données de respiration cellulaires, mitochondriales, qui peuvent être
\ngénérées grâce à l'utilisation d'une plateforme d'étude métabolique, le Seahorse.</br> <br>T lymphocytes are important components of the adaptive immune response in the elimination of
\nvarious infections. ln the case of viral infections, this elimination takes place via the production of
\nantiviral cytokines by the helper T-CD4s to activate the various immune populations necessary,
\nas well as by the T-CD8s which are also known for their cytolytic action leading directly to lysis of
\nthe infected cell.</br> <br>
\nln order to ensure optimal antiviral and cytotoxic responses, lymphocytes require a significant
\nenergy supply to proliferate and express several cytokines simultaneously, hence the term
\npolyfunctionality. This energy supply is ensured by metabolic plasticity allowing cells to alter their
\nmetabolic pathways in order to degrade various metabolites and transform them into ATP. Several
\nstudies, including those conducted in our laboratory, show that one of the pathways most used
\nby activated T lymphocytes is fatty acid beta oxidation (FAO).</br> <br>
\nSeveral substances can inhibit cellular immune responses, such as cannabinoids, the
\nconsumption of which is legal in Canada. Cannabis consumption by the general population is
\npredominantly full-spectrum cannabis with either high cannabidiol (CBD) content or high
\ntetrahydrocannabinol (THC) content. The effect of CBD is rather therapeutic since it is known for
\nits anti-inflammatory and pain-relieving effect. However, recreational Cannabis users seek a
\nneurotropic effect referred to as 'High', which is typically found when consuming cannabis with a
\nhigh THC content. This consumption, having increased since the legalization of cannabis, and all
\nthe more since the beginning of the current pandemic in order to reduce mental stress due to the
\nconditions of isolation and distancing imposed in Quebec. </br> <br>
\nln the context of the need to generate a sufficient and protective immune response against SARSCoV-
\n2 following vaccination, one wonders if this consumption of THC wou ld not be of an inhibiting
\nnature with regard to the vaccine response of consumers. recreational workers, who represent a
\nfairly la rge population in Quebec and Canada.</br> <br>
\nAs a result, there has been interest in studying the effect of THC on the antiviral responses of
\nmemory T-CD8s and T-CD4s.</br> <br>
\nThis study was mainly carried out using the flow cytometry platform allowing us to target from
\nperipheral blood mononuclear cells (PBMCs), our cells and cytokines of interest, being respectively T-CD8s lymphocytes and IFNy, TNFa as well as IL-2, via a multiparametric cocktail
\nof antibodies. Our results confirmed that T-CD8s lymphocytes, polyclonally activated and, treated
\nor not, with THC for 24 hours, show a significant inhibition of their immune polyfunctionality. These
\ncells also show an inhibition of their cytotoxic potential observed via their expression of the cellular
\ncytotoxicity molecules, Granzyme B and Perforin . lmportantly, using the same experimental
\nplatform, we demonstrate a possible impairment of lipid metabolism, by evaluating the intramitochondrial
\nexpression of the lipid transporter CPT1 a, which is significantly reduced in T-CD8s
\nlymphocytes activated in the presence of THC. These primary metabolic data must be confirmed
\nwith cellular and mitochondrial respiration data, which can be generated through the use of a
\nmetabolic study platform, the Seahorse. </br>
