Abstract
Chemicals. HPLC grade organic solvents including methanol, acetonitrile, chloroform and methylene chloride were purchased from Sigma/Aldrich (St. Louis, MO). N-Palmitoylethanolamine (PEA), N-stearoylethanolamine (SEA), heptadecanoyl chloride, triethylamine and butylated hydroxytoluene (BHT) were also purchased from Sigma/Aldrich (St. Louis, MO). Ethanol-1,1,2,2-d4-amine (99.5 % D) was purchased from CDN Isotopes Inc (Quebec, Canada). 1,2-Dipalmitoyl-sn-glycero-3-phosphoethanolamine was purchased from Avanti Polar Lipids, Inc. (Alabaster, AL). The water used is generated in-house with 18 megaohm-cm resistivity using a Millipore water purification system. Lipid Extraction from Plasma and Lymph Samples for LC/MS/MS Analyses. Plasma samples (100 μL) were mixed with 2.0 ml pre-chilled chloroform/methanol (v/v, 2:1, containing 0.01 % BHT) in a glass vial, then combined with C17-d4 NAE, C17 NAPE and C17C17 PE internal standard solutions and 250 μl water. The mixtures were vortexed for 30 seconds before centrifugation at 4000 rpm (4 o C) for 20 min. The bottom organic phase was carefully collected and dried with a steady stream of nitrogen gas.
