Historical biogeography and conversation of eastern pipistrelles (Pipistrellus subflavus) in Nova Scotia

Abstract
The eastern pipistrelle, Pipistrellus subflavus, is a species of bat found throughout eastern Canada, United States (US), and Mexico. In Atlantic Canada the only known population is found in southwestern Nova Scotia (NS). The population found in and around Kejimkujik National Park, NS appears morphologically and behaviourally distinct from other populations. Furthermore this population is likely genetically isolated from populations in the US and the rest of Canada. The present study aims to compare cytochrome b gene sequences from the NS population of Pipistrellus subflavus with other populations in Canada and the US to determine whether they warrant recognition as an evolutionary significant unit or even a distinct species. Standard methods of DNA isolation from small (<1 mg) skin samples resulted in DNA isolations of low concentration and contamination with PCR inhibitors. A Chelex-based DNA isolation method appeared to remove or reduce the PCR inhibitor(s) but DNA concentrations continued to be very low. Nevertheless, a 400 base pair segment of the cytochrome 6 gene was successfully amplified and sequenced for eight specimens and these were compared with sequences from five P. subflavus specimens downloaded from the Genbank database. Sequences were compared using traditional phylogenetic analyses. This study showed <2% sequence variation in this portion of the cytochrome 6 gene among populations of Pipistrellus subflavus, which is consistent with traditional levels of intraspecific variation in bats. These findings suggest that the behavioural and morphological differences that have been documented may be a result of local environmental factors influencing growth and roosting habits rather than underlying genetic differences, This study provides the first look at a potential genetic basis for differences seen in the disjunct NS population of Pipistrellus subflavus in comparison to other populations.
