La synthèse des protéines est un processus qui intègre divers signaux intracellulaires et extracellulaires, notamment la disponibilité des acides aminés, le stress et la présence de facteurs de croissance. La régulation de la traduction façonne le protéome cellulaire et est essentielle à l'homéostasie. Dans le cerveau, la formation de la mémoire à long terme (MLT) est un processus dépendant de la synthèse des protéines. Il a été démontré que les changements dans la synthèse des protéines modifient la MLT. La voie de signalisation PI3K/Akt/mTOR, qui régule l'initiation de la traduction, a été associée à la mémoire et à l'apprentissage. L'une des cibles de phosphorylation en aval de mTORC1 (mechanistic target of rapamycin complex 1) est 4E-BP (eIF4E-binding protein). Il existe trois paralogues de cette protéine (4E-BP1, 4E-BP2, 4E-BP3), mais seuls les deux premiers sont présents en quantité significative dans le cerveau, 4E-BP2 étant l'isoforme la plus répandue. Le lien entre 4E-BP2 et la formation de la MLT a été largement étudié. Chez la souris, la délétion de 4E-BP2 au niveau du corps entier entraîne une altération de la mémoire. On a découvert que les neurones inhibiteurs étaient responsables de ce phénotype. Cela montre qu'une synthèse protéique excessive peut être préjudiciable, soulignant la nécessité d'une régulation étroite pendant la formation de la mémoire. Le rôle de l'isoforme 4E-BP1 n'est cependant pas clair dans le contexte de la formation de la MLT. Nous avons donc cherché à étudier son rôle dans la mémoire. Nous montrons que la délétion germinale du gène codant pour 4E-BP1 chez les souris C57BL/6 n'altère pas la localisation/reconnaissance d'objets, la mémoire de travail ou la mémoire spatiale, mais qu’elle augmente la mémoire de la peur contextuelle. De plus, la délétion conditionnelle de 4E-BP1 dans les neurones inhibiteurs exprimant l’acide glutamique décarboxylase 2 (Gad2), mais pas dans les neurones excitateurs exprimant la sous-unité alpha de la protéine kinase calcium/calmodulin-dépendante de type II (CamKIIα), entraîne des augmentations similaires de la mémoire de peur contextuelle. En outre, la délétion de 4E-BP1 augmente l'expression de PSD95, une protéine post-synaptique importante pour la formation de la MLT. L'ensemble de ces résultats suggère que l’invalidation génique de 4E-BP1 améliore la mémoire de peur à long terme par l'intermédiaire des neurones inhibiteurs et de l'expression des marqueurs post-synaptiques
