Abstract
Translational regulation mechanism is crucial for proper protein synthesis and mRNA quality control during developmental and cellular processing.Regulation of mature mRNA would reflect faster on protein levels compared to transcription controls.Protein synthesis can be regulated at all stages, namely initiation, elongation, termination, and ribosome recycling.However, most regulation occurs in the initiation step.mRNA translation initiation repression can be mediated through different eIF4E-homologous protein (4EHP) -(GRB10 interacting GYF protein 2) GIGYF2 containing complexes.ZNF598 binds to GIGYF1/2 GYF domain with Cterminal polyproline motif, and the complex could repress translation of inflammatory response genes under resting state.The non-structural protein 2 of SARS-CoV-2 can interact with the host's GIGYF2 and enhance the interferon beta repression through 4EHP-GIGYF2 dependent fashion, which results in repression of the innate immune response and inhibition of antiviral gene activation.So far, the structure of GIGYF2, ZNF598, and NSP2 has not been resolved yet.This thesis focuses on the expression and purification of these proteins and aiming for structural study of 4EHP-GIGYF2-ZNF598 complex with cryo-EM and that of NSP2 with X-ray Crystallography.professionally-have greatly contributed to my academic growth.My passion for structural biology was sparked and nurtured under his guidance, and for that, I am profoundly grateful.
