La maladie de Parkinson est parmi les maladies neurodégénératives les plus répandues. Elle se caractérise par la perte des neurones dopaminergique dans le mésencéphale. Bien que la majorité des cas soit sporadique, environ 10% des patients ont une forme familiale de la maladie. Des mutations pathogènes de PRKN ou PINK1 cause la maladie de Parkinson à début précoce. La voie PINK1/parkin est responsable de l’élimination des mitochondries endommagées via mitophagie. PINK1 est une kinase qui régule l’activité de la E3 ligase parkin assurant que parkin ne devient actif qu’à la suite de dégâts aux mitochondries. Malgré le progrès dans les études structurales et biochimiques de parkin, deux domaines de recherche restent insuffisamment compris : la pathogénicité de certains variants de parkin et la sélectivité pour ses substrats. Ici, j’ai caractérisé deux mutations dans le domaine catalytique, T415N and P437L, et j’ai étudié l’interaction de parkin avec son substrat préféré mitofusin 2 (Mfn2) utilisant une combinaison des essais biochimiques et biophysiques avec de la modélisation par AlphaFold. Cette approche a révélé un mécanisme structural pour la perte de fonction des mutations dans le domaine catalytique de parkin, attribué à la perturbation de la liaison de l’ubiquitin. J’ai identifié une α-hélice non observée auparavant qui positionne l’ubiquitine pour le transfert de la E2 à parkin. Pour étudier la sélectivité de parkin pour ses substrats, j’ai modélisé le complexe formé par parkin activé et Mfn2 qui met en évidence des interactions importantes entre le domaine catalytique de parkin et le domaine hélical de Mfn2. J’ai ensuite conçu et purifié des constructions de Mfn2 soluble conservant les sites d’ubiquitination ce qui sera utile pour des études in vitro. Les résultats contribuent vers une meilleure compréhension de la biologie de parkin et ouvrent la voie pour le développement de nouveaux thérapeutiques
